A time series analysis of bulk tank somatic cell counts of dairy herds located in Brazil and the United States Ciência Rural
Rodrigues,Liz Gonçalves; Aquino,Maria Helena Cosendey de; Silva,Márcio Roberto; Mendonça,Letícia Caldas; Mendonça,Juliana França Monteiro de; Souza,Guilherme Nunes de.
ABSTRACT: Bulk tank somatic cell counts (BTSCC) is widely used to monitore the mammary gland health at the herd and regional level. The BTSCC time series from specific regions or countries can be used to compare the mammary gland health and estimate the trend of subclinical mastitis at the regional level. Three time series of BTSCC from dairy herds located in the USA and the Southeastern Brazil were evaluated from 1995 to 2014. Descriptive statistics and a linear regression model were used to evaluate the data of the BTSCC time series. The mean of annual geometric mean of BTSCC (AGM) and the percentage of dairy herds with a BTSCC greater than 400,000 cells mL-1 (%>400) were significantly different (P<0.05) according to the countries and the times...

Antibiotic activity of Plectranthus ornatus Codd., a Traditional Medicinal Plant Anais da ABC (AABC)
NASCIMENTO,FERNANDA R.; ALBUQUERQUE,KAMYLLA R.S.; OLIVEIRA,MARCOS R.; PIZZIOLO,VIRGINIA R.; BRASILEIRO,BEATRIZ G.; DIAZ,GASPAR; DIAZ,MARISA A.N..
ABSTRACT The dichloromethane extract of Plectranthus ornatus Codd., a tradicional medicinal plant, showed antibiotic activity with minimum inhibitory concentration (MIC) values of 0.4 mg.mL-1 and 100 percent of biofilm inhibition against Staphylococcus aureus strains isolated from animals with mastitis infections. Based on these antibacterial activities, in addition to ethnopharmacological reports from healing men and farmers in Brazil, an herbal soap was produced from this active extract and was tested both in vitro and in vivo. In vivo assays conducted on these herbal soaps led to results similar to those previously conducted with the active extract. These results indicated the great potential of this plant for use as an excipient by preparing herbal...

Bovine S protein (vitronectin) increases phagocytosis of Streptococcus dysgalactiae Rev. Microbiol.
Filippsen,Laerte Francisco.
The effects of bovine S protein (vitronectin) on phagocytosis of Streptococcus dysgalactiae strains isolated from cattle with mastitis were investigated. Phagocytized streptococci were determined by a fluorometric microassay using glass adherent polymorphonuclear neutrophils (PMN). Preincubation of S. dysgalactiae with bovine S protein significantly increased their phagocytosis by PMN. Bovine S protein had no effect on phagocytic killing of non-S protein binding S. pyogenes cultures. Enzymatic digestion of the bovine S protein binding sites on S. dysgalactiae with pronase resulted in a significative reduction of the effects of S protein on phagocytosis. It could thus be concluded that in addition to its role as a promoter of cellular adhesion and...

Comparison of methods for the detection of biofilm formation by Staphylococcus aureus isolated from bovine subclinical mastitis BJM
Melo,Poliana de Castro; Ferreira,Luciano Menezes; Nader Filho,Antônio; Zafalon,Luiz Francisco; Vicente,Hinig Isa Godoy; Souza,Viviane de.
Biofilm formation is considered to be a selective advantage for Staphylococcus aureus mastitis isolates by facilitating bacterial persistence in the udder. It requires attachment to mammary epithelium, proliferation and accumulation of cells in multilayers. The objective of this study was to determine the sensitivity and specificity of three techniques for the detection of S. aureus biofilm-positive strains. Two phenotypic tests, including growth on microtitre plates and Congo red agar, were compared with a PCR technique using 94 S. aureus strains obtained from cows with subclinical mastitis from two farms in the state of São Paulo. These strains were characterised by in vitro slime production on Congo red agar, biofilm formation on microtitre plates and...

Detection of different Staphylococcus aureus strains in bovine milk from subclinical mastitis using PCR and routine techniques BJM
Vieira-da-Motta,Olney; Folly,Márcio Manhães; Sakyiama,Cássia Camargo Hagen.
Contamination of fresh milk with Staphylococcus aureus was assessed comparatively through routine phenotypic (coagulase tube test and coagulase slide test) and genotypic (PCR) screening of 128 S. aureus strains isolated from 555 milk samples. These samples were collected from 362 cows with subclinical mastitis, hosted in different dairy herds at various locations of the Northern and Northeastern rural areas of the State of Rio de Janeiro, 39.7% of which were CMT-positive. All S. aureus isolates tested positive for the presence of the coagulase gene by PCR and the isolates could be grouped into four distinct classes according to the size of the PCR product. The strains also yielded variable results when assayed with coagulase test. Taken together, these...

Melo,Dayanne Araújo de; Coelho,Irene da Silva; Motta,Cássia Couto da; Rojas,Anna Carolina Coelho Marín; Dubenczuk,Felipe Carlos; Coelho,Shana de Mattos de Oliveira; Souza,Miliane Moreira Soares de.
Staphylococcus aureus antimicrobial resistance, especially to beta-lactams, favors treatment failures and its persistence in herd environment. This work aimed to develop a more specific primer for mecA gene detection based on the comparison of the conserved regions from distinct host origins and also investigated the presence of homologue mecA LGA251 in bovine strains. A total of 43 Staphylococcus spp. were included in this study, comprising 38 bovine S. aureus, two human and three equine coagulase-negative staphylococci (CNS). Phenotypical methicillin-resistance detection was performed through oxacillin agar-screening and cefoxitin disk-diffusion test. None isolate tested positive for mecA LGA251 gene. For mecA gene PCR, new primers were designed based on...

Isolation and identification of main mastitis pathogens in Mexico Arq. Bras. Med. Vet. Zootec.
Castañeda Vázquez,H.; Jäger,S.; Wolter,W.; Zschöck,M.; Castañeda Vazquez,M.A.; El-Sayed,A..
The present work is a large epidemiological study aiming to detect the prevalence of subclinical mastitis and to investigate the major udder pathogens in Jalisco State, western Mexico. For this purpose, 2205 dairy cows, representing 33 Mexican dairy herds, were involved. Of 2205 cows, 752 mastitic animals were diagnosed and only 2,979 milk samples could be obtained for further investigation. All 2979 milk samples were subjected to California Mastitis Test (CMT) to differentiate clinical cases from subclinical ones where 1996 samples (67 %) reacted positively. Of these, 1087 samples (54.5%) came from cows suffering from clinical cases of mastitis. Bacteriological identification of the causative agents revealed the presence of a major group of pathogens...

Plant Extracts Display Synergism with Different Classes of Antibiotics Anais da ABC (AABC)
SILVA,DANIELLE M.; COSTA,PRISCILLA A. DA; RIBON,ANDRÉA O.B.; PURGATO,GISLAINE A.; GASPAR,DIAZ-MUÑOZ; DIAZ,MARISA A.N..
Abstract: One manner in which plant-derived compounds exert their antibiotic potential is the synergism, a positive interaction between two compounds. Studies indicate that the use of plant extracts combined with antimicrobials may promote a significant reduction of the minimum inhibitory concentrations of antibiotics for bacterial strains. This study aimed to evaluate the activity of plant extracts and antibiotics as well as their combination on Staphylococcus aureus. The activity of 15 plant extracts was evaluated using diffusion assay. The minimum inhibitory concentrations (MICs) and the interactions between the extracts and antibiotics as well as compound emodin were evaluated with the checkerboard method. The active extracts were a hexane extract of...

Resistência bacteriana e ação das bacteriocinas de Lactobacillus spp em Staphylococcus aureus isolados de mastite bovina Arq. Bras. Med. Vet. Zootec.
Pribul,B.R.; Pereira,I.A.; Soares,L.C.; Coelho,S.M.O.; Barberis,I.L.; Pascual,L.; Souza,M.M.S..
Staphylococcus aureus is the major pathogen causing intramammary infections in dairy cattle worldwide. Among the factors that contribute to its spread and infectious potential is the ability to overcome the mechanisms of antimicrobials activity. The present work investigated the antimicrobial resistance pattern and sensibility to bacteriocins produced by strains of Lactobacillus spp of 30 isolates of S. aureus from mastitis. From this, 29 are beta-lactamase producers. Eight isolates (26.6%) showed resistance to at least four antibiotics being considered multiresistent. All of them were mecA-positive. Otherwise, all isolates tested showed sensibility to at least one of the four bacteriocin producer strains. Due to the significant depletion of the efficacy...

The Matrix-Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry (MALDI-TOF MS) identification versus biochemical tests: a study with enterobacteria from a dairy cattle environment BJM
Rodrigues,Naiara Miranda Bento; Bronzato,Greiciane França; Santiago,Gabrielli Stefaninni; Botelho,Larissa Alvarenga Batista; Moreira,Beatriz Meurer; Coelho,Irene da Silva; Souza,Miliane Moreira Soares de; Coelho,Shana de Mattos de Oliveira.
Abstract Mastitis adversely affects milk production and in general cows do not regain their full production levels post recovery, leading to considerable economic losses. Moreover the percentage decrease in milk production depends on the specific pathogen that caused the infection and enterobacteria are responsible for this greater reduction. Phenotypic tests are among the currently available methods used worldwide to identify enterobacteria; however they tend to misdiagnose the species despite the multiple tests carried out. On the other hand The Matrix-Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry (MALDI-TOF MS) technique has been attracting attention for its precise identification of several microorganisms at species level. In...
